Creation of file sound works. But i fi i open it and run each command manually i have an error.

If you're doing it manually, you'll need to prefix each of those commands with sudo.

i think is impossible to ear differences. Is not easy to switch configuration for a A/B test

does this tweak improve the sound via Hat Dac RCA out?

or is the tweak only for usb out

It's not just for USB output.

Try it and see if you can hear a difference.
