I would stick to Samba as the protocol between your server and your player. It works just fine.
Tim has added some very nice functionality to moOde by integrating miniDLNA and djmount, but unless there is other UPnP/DLNA-only equipment involved, it isn't necessary to use this functionality. IMHO the question isn't "does UPnP work better?", it's "is UPnP necessary?".
Of course, the host with your big drive can be configured to speak more than one protocol. The only cost is the configuration effort and the CPU/network overhead.
